The Celloidin Embedding and Routine Staining Method



Abstract
The tissue are damaged by heated paraffin and a room ternperature embedding medium must be used, the choice would be celloidin and functional elements which go to make up the nervous system. There are many special techniques capable of demonstrating nervous tissue entities. The greatest risk to successful embedding is rater contamination. The requisite for successful infiltration of tissue is perfect dehydration with absolute alcohol that must be kept in airtight containers to avoid water from atmospheric conditions. The ether must be anhydrous, not comr nercial ether. The dehydration must be done in airtight containers, starting at low-grade alcohols and ending with absolute alcohol-ether 1 : 1. Clearing agents are not used in this method. The process may require weeks or even months. infiltration is dor_e in graded solutions of , 8;0, and 1200 celloidir-. The time will veary with the size and nature of the tissue. Celloidin sections are usually stared in small slender dishes or % itch glasses and handled by being moved from one fluid to ane_aer with a section lifter. The celloidin protects the section against damage.
